AffiPro offers a set of five different types of acidic protease columns for on-line digestion with different cleavage preferences:

- Rhisopuspepsin
- Nepenthesin-1
- Nepenthesin-2
- Aspergillopepsin
In 2022 AffiPro also introduced a new column with mixed stationary phase of Nepenthesin-2/Pepsin combination specifically optimized for antibodies and membrane proteins.
Optimization of protein digestion is a key step in the overall HDX experiment. Spatial resolution and complete protein sequence coverage are the crucial parameters necessary for obtaining useful structural information.
The selection of protease column or their combination together with smart choice of denaturant agents and flow rate boosts digestion performance and allows to achieve full coverage of the protein sequence with many overlapping peptides.
